Zeocin? Reagent is a member of the bleomycin antibiotic family. Resistance is conferred by the Sh ble gene which encodes a 13,665 dalton protein. In cells expressing the protein, Zeocin? is prevented from binding and cleaving the cellular DNA. Zeocin? is effective on most aerobic cells and can be used for selection in mammalian and insect cell lines, yeast, and bacteria. The amount of Zeocin? required for selection will vary between 50-2000 μg/ml (typically 300μg /ml) depending on the cell type.

Zeocin可選擇Sh ble基因表達(dá)的細(xì)胞。與現(xiàn)在使用的其他動(dòng)物細(xì)胞標(biāo)記物無(wú)交叉抗性。因此這種抗生素可用來(lái)分離對(duì)其他篩選劑(如:慶大霉素,潮霉素)有抗性的克隆。Zeocin是一種屬于爭(zhēng)光霉素家族的糖蛋白抗生素,在體內(nèi)能作用于大多數(shù)細(xì)菌(包括E. coli)、真菌(如:酵母菌)、植物細(xì)胞、動(dòng)物細(xì)胞。

● 液體Zeocin可用于細(xì)胞培養(yǎng)。

● 用于篩選轉(zhuǎn)化株的Zeocin濃度根據(jù)pH值和鹽濃度改變。pH越高,鹽濃度越低,Zeocin活性越大。

● 普通大腸桿菌菌株篩選可在25 μg/ml zeocin濃度的低鹽LB瓊脂培養(yǎng)基(yeast extract 5 g/L, Tryptone 10 g/L, NaCl 5 g/L, Agar 15 g/L,pH 7.5)中進(jìn)行,pH調(diào)至7.5 。

● 動(dòng)物細(xì)胞篩選:根據(jù)不同細(xì)胞系從100 到 750 μg/ml。

● 植物細(xì)胞篩選:根據(jù)不同的細(xì)胞種類和培養(yǎng)基從50 到 250 μg/ml 。

● 酵母篩選:取決于種屬和培養(yǎng)基,S. cerevisiae Sh ble轉(zhuǎn)化株在50 μg/ml 濃度的YPD培養(yǎng)基中篩選,pH調(diào)至7.0。

粉末型配制方法:

取1 ml Zeocin溶解液加到Zeocin干粉中,徹底溶解,用0.22 μm 一次性濾器抽濾除菌,即配成1 ml(100mg/ml)的Zeocin溶液,-20℃保存。
